ABSTRACT

Objective To analyze the profile of dermcidin (DCD) changes in different stages of acute coronary syndrome (ACS) by quantifying the serum 4 183Da DCD peptide fragment deriving from different ACS patients treated with early antithrombotic therapy. Methods A total of 118 patients with confirmed diagnosis of ACS were enrolled. Immediately after visiting a doctor, the venous blood was collected and afterwards instantly the patient was given orally 300 mg of aspirin and 300 mg clopidogrel, and according to the patient's condition and the consent of his/her or acknowledgement of family members achieved, emergency percutaneous coronary interference (PCI) or thrombolysis or conservative treatment was adopted separately. After anti-thrombotic treatment, at 2, 4, 6, 8, 10, 12, 16, 20, 24, 32, 40, 48, 60 and 72 hours, venous blood was collected and serum isolated respectively. The concentration of 4 183Da DCD fragment in serum was determined by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). Simultaneously, the myoglobin (Myo), cardiac troponin I (cTnI) and MB isoenzyme of creatine kinase (CK-MB) were also detected. Results The mean relative strength of nature logarithmic transformations of 4 183Da DCD fragment of 118 patients with ACS was 2.75±1.02 before treatment on admission, and after intervention therapy (mainly antithrombotic therapy) it was decreased to 1.84±1.19 (P = 0.005) and 1.74±1.12 (P = 0.000) at 2 hours and 4 hours, respectively, and then after 4 hours it was slightly elevated. 4 183Da polypeptide increased earlier than myocardial injury markers. Conclusion Aspirin and clopidogrel can significantly decrease the concentration of 4 183Da DCD peptide fragment in serum in patients with ACS, which indicates that the DCD fragment could be used as one of the indexes for observation on early efficacy of antithrombotic therapy.

ABSTRACT

Objective To observe the effect and mechanism of HA1077 on bone mesenchymal stem cells (BMSCs) promoting wound healing. Methods (1) In vitro study: rats' BMSCs were har-vested and divided into three groups randomly, ie, control group, induction group and HA1077 group. In control group, culture medium was DMEM with 10% fetal bovine serum. In induction group, induction medium was DMEM which consisted of 10% FBS, 8μg/ml EGF and epithelium culture condition medi-um. In HA1077 group, culture condition medium was the induction medium pins 10 μmol/L HA1077. At day 7 after replanting, CK19 expression, PCNA expression and cell cycle among three groups were clarified by flow cytometry. (2) In vivo study: rabbit's BMSCs were isolated. Six pieces of full skin loss wound were created on the back of a rabbit, with three pieces of wound on the half side of back. The wound was round and 3 cm in diameter. Eight New Zealand rabbits were randomly divided into four groups, ie, control group (Group A), EGF control group (Group B), BMSCs group (Group C) and BMSCs + HA1077 group (Group D). Wounds were treated by normal saline in Group A and by EGF (50 U/cm2) in Group B. Besides EGF,1 ml of BMSCs suspension (2 × 106/ml) was used for wound re-pair in Group C. 0.5 ml HAIO77(20 μmol/L), BMSCs and EGF were used together for wound repair in GronpD. At days3, 7and 14 after injury, wound elosuroindex (WCI) was calculated. Results In vitro study:the positive rate of CK19 in the HA1077 group was (40.31±0.82)%, higher than (11.13±2.14)% in the induction group(P < 0.01). The PCNA positive rate and the S phase, cell quantity in HA1077 group were also higher than that in the induction group. In vivo study: WCI was (86.20±1.92) % in Group D at day 14 after injury, obviously higher than that in other three groups. Conclusions HA1077 can promote BMSCs' entering into S phase, expressing PCNA and differentiating and expressing CK19. In vivo combination of HA1077 with BMSCs may facilitate wound healing.

ABSTRACT

OBJECTIVE To reconstruct the HNP-1 into the J-HNP-1 with a J chain, and explore to set up a mammalian cell expression system which can express and secret J-HNP-1,so that the products could be examined and purified conveniently. METHODS The J-HNP-1 cDNA fragments were produced by recombinant PCR.Then the J-HNP-1 was inserted into the mammalian expression vector pcDNA3.1(-)/Myc-His which had the double marks Myc and 6?His.The recombinant vector rpcDNA3.1(-)/Myc-His /J-HNP-1 was transfected into the COS-7 cell. The J-HNP-1 expression was analyzed at the mRNA and protein level. The germicidal activity of cell culture supernatant and cellular solution protein was assayed. RESULTS By the use of RT-PCR with special primers,a band of 786bp was amplified from COS-7 cells transfected by this recombinant plasmid.Western blot analysis with specific anti-histidines antibody revealed that the cell culture supernatant and cellular solution protein of COS-7 cells transfected by rpcDNA3.1(-)/Myc-His /J-HNP-1 had a strong band with relative molecular mass of about 24?10~3.Antibacterial activity assay showed that obvious bacterial inhibition occurred in both lysate and supernatant of COS-7 cells transfected by rpcDNA3.1(-)/Myc-His /J-HNP-1. CONCLUSIONS The J-HNP-1 recombinant is obtained and inserted into the mammalian expression vector then to be expressed in vitro. The expression product is found to have the anti-bacterial effect in vitro.

Totally 300 home-going peasant-workers weresurveyed during the Spring Festival.METHODS: The peasant-workers were tested individually with the Satisfaction with Life Scale (SWLS). It was made up of 5 items:The first item was "In most ways my life is close to my ideal ", the second was "The conditions of my life are excellent", the third was "I am satisfied with my life", the fourth was "So far I have gotten the important things I want in life", the fifth was "If I could live my life over, I would change almost nothing". Using the 1-7 scale below: 1-Strongly disagree, 2-Disagree, 3-Slightly disagree, 4-Neither agree nor disagree, 5-Slightly agree, 6-Agree, 7-Strongly agree. The test-retest reliability of this scale was higher than 0.80, the content-related validity was 0.60, and the crieterionrelated validity was higher than 0.50. They were given uniform directions and answered each question individually according to their own conditions in the latest week. All the answer papers were handed in on the spot. The data of SWLS were input into the computer for statistical analysis. The questionnaire in which more than 4 items were selected the answer of "neither agree nor disagree" were considered to be invalid and therefore rejected for analysis. All the valid data were input into the computer and statistically analysed with SPSS 11.0 software; differences among groups were compared with t test or analysis of variance.MAIN OUTCOME MESURES: The scores of SWLS answered by the peasant-workers.RESULTS: Totally 300 questionnaires were distriluted and 275 of them were collected, with a recovery rate of 91.67%, of which 245 were valid for result analysis, including 118 males and 127 females aged from 16 to 52 years old with the mean age of 26.09 years. They came from 15 provinces of China and from 16 kinds of vocation. Their monthly-earning was from 200 yuan to 8 000yuan. The investigated results of SWLS showed that: ①29.8% of the tested home-going peasant-workers were inclined to be satisfied with their life, and 41.6% of them were inclined to be unsatisfied. ②There was obviously sexual difference in the item of "I am satisfied with my life"among the tested homegoing peasant-workers.[(4.50±1.70,3.95±1.53) (t=2.65,P < 0.001 )]. ③Earning markedly affected their feeling of "The conditions of my life are excellent" and "So far I have gotten the important things I want in life" [(3.79±1.85, 3.98±1.64,4.07±1.44,4.84±1.14) (F=3.068,P < 0.05); (4.64±2.03,3.72±1.68,4.07±1.67,4.19±1.28) (F=2.813,P < 0.05)]. ④there was no significant difference of satisfaction with life of the tested home-going peasant-workers in educational level and term on duty. CONCLUSION: Majority of the home-going peasant-workers were unsatisfied with their living. Sex and earning were the important factors that affected their satisfaction with life, educational level and term on duty had weakly correlated with it.
